Molybdenum and Nitrate Reductase

چکیده

Numerous investigators have shown that nitrate nitrogen accumulates in higher plants (l-3) and fungi (2, 4) which are deficient in molybdenum. Other workers have shown that sulfur, zinc, and manganese deficiencies in higher plants may also result in a similar phenomenon (5). The free amino acids present in molybdenum-deficient plants are usually less than those in normal tissues, thus indicating that molybdenum is important for the metabolism of nitrogen (2.-4). The function of molybdenum in enzyme systems involved in the utilization of nitrogen has not hitherto been established. The characterization of nitrate reductase, including evidence for the presence of a metal component, and the demonstration that iron, manganese, and zinc deficiencies actually increased the specific activity of the enzyme (6) prompted an investigation of the effect of molybdenum deficiency on nitrate reductase. This paper elaborates a preliminary report (7) in which it was shown that molybdenum is specifically required in the nutrient medium for the activity of nitrate reductase in the fungi Neurospora crassa and Aspergillus niger.

Role of molybdenum in nitrate reduction by chlorella.

Molybdenum is absolutely required for the nitrate-reducing activity of the nicotinamide adenine dinucleotide nitrate reductase complex isolated from Chlorella fusca. The whole enzyme nicotinamide adenine dinucleotide nitrate reductase is formed by cells grown in the absence of added molybdate, but only its first activity (nicotinamide adenine dinucleotide diaphorase) is functional. The second a...

Electron Paramagnetic Resonance Studies on the Molybdenum Center of Assimilatory NADH : Nitrate Reductase from ChZoreZZu uuZguris

The assimilatory nitrate reductase from Chlorella contains flavin, heme, and molybdenum as prosthetic groups. The molybdenum in assimilatory nitrate reductase is associated with a pterin moiety (molybdopterin) as evidenced by the ability of the enzyme to donate active molybdenum cofactor to the Neurospora nitrate reductase mutant nit-1 and by the oxidative conversion of the pterin to two well c...

Invitro formation of assimilatory reduced nicotinamide adenine dinucleotide phosphate: nitrate reductase from a Neurospora mutant and a component of molybdenum-enzymes.

An active Neurospora-like assimilatory NADPH-nitrate reductase (EC 1.6.6.2), which can be formed in vitro by incubation of extracts of nitrate-induced Neurospora crassa mutant nit-1 with extracts of (a) certain other nonallelic nitrate reductase mutants, (b) uninduced wild type, or (c) xanthine oxidizing and liver aldehyde-oxidase systems was also formed by combination of the nit-1 extract with...

Characterization of molybdenum cofactor from Escherichia coli.

Molybdenum cofactor activity was found in the soluble fraction of cell-free extracts of Escherichia coli grown aerobically in media supplemented with molybdate. Cofactor was detected by its ability to complement the nitrate reductase-deficient mutant of Neurospora crossa, nit-1, resulting in the vitro formation of nitrate reductase activity. Acid treatment of E. coli extracts was not required f...
